RESUMO

Enzyme immobilization provides ideal operating conditions for enzymes stabilization and sustainable recycling. In this work, as a kind of clay material, montmorillonite (MTL) was chosen for immobilizing the ß-glucosidase extracted from Agrocybe aegirit. The immobilized ß-glucosidase via partly cross-linking enzyme aggregates (pCLEAs) formed by self-catalysis provided biocatalysts with satisfactory thermal and pH stability. Compared to the glutaraldehyde cross-linked, the immobilized ß-glucosidase (ß-G-pCLEAs@MTL) exhibited significantly higher immobilization efficiency (IE) and immobilization yield (IY), which were 80.6% and 76.9%, respectively. The ß-G-pCLEAs@MTL also showed better stability and preferable reusability. And the activity of the ß-G-pCLEAs@MTL remained 85.0% after 5 cycles and 74.7% after 10 cycles. Therefore, the method based on the pre- crosslinking to form pCLEAs and after-immobilization can effectively improve IY and IE. In addition, MTL seems to be a good alternative carrier to immobilize other enzymes for industrial application.

N-butylimidazolium functionalized strongly basic anion exchange resin with Cl(-) anion (MCl) was prepared by anchoring N-butylimidazole onto chloromethylated macroporous styrene-divinylbenzene (St-DVB) copolymer. The adsorption performances of phenol on MCl were studied using the batch technique at acidic and alkaline pH. The studies showed that phenol can be effectively removed at both acidic and alkaline pH. The maximum adsorption was achieved at about pH 11. The maximum adsorption capacities of phenol on MCl at pH 6.6 and 11.2 were 80.2 and 92.9 mg/g, respectively. The adsorption mechanism was mainly molecular adsorption at acidic pH and anion exchange at alkaline pH. The adsorption of phenol was hindered by the presence of Cl(-) and SO(4)(2-) at alkaline pH due to the competitive anion exchange reaction. The adsorption of molecular phenol species on MCl at acidic pH was exothermic, and the anion exchange of phenolate species by MCl at alkaline pH was endothermic. Desorption of phenol from loaded adsorbent was achieved by using 0.5 mol/L NaOH and 0.5 mol/L NaCl mixed solution. MCl can simultaneously remove phenol and Cr(VI) from their mixtures, which would be of practical value in actual industrial wastewater treatment.

Acclimatized the activated sludge with [omim][PF6] which was used as the C, N, P resources for microbe, the biodegradation ratio of [omim][PF6] with different inoculants which were acclimatized or not was investigated. And the influence of appending an ester in the side chain of the imidazolium cation on the biodegradation of [omim][PF6] was discussed in the same conditions. The biodegradation pathways of [omim]+ were analyzed. The results showed that the biodegradation of ionic liquids was improved by appending an ester in the side chain; the biodegradation rate of [omim][PF6] was less than 20% when the inoculant was original activated sludge, which was not referred as "readily biodegradable"; [omim][PF6] would stay in environment for a long time; the biodegradation ratio reached 60% when the inoculant was acclimatized activated sludge; 1-methyl-imidazolium was residual in the course of biodegradation of [omim]+, the semifinished products of biodegradation of [omim]+ was analyzed by HPLC-MS/MS and the pathways were presumed.

Malignant angiomyolipoma (AML) of the liver is rare. We report a case of AML with malignant transformation and metastases. A 30-year-old man had developed giant hepatic masses. Microscopically, the periphery of the tumor showed components of classic hepatic AML, but the central region contained atypical epithelioid components with extremely pleomorphic and hyperchromatic nuclei with frequent mitotic figures. Immunohistochemical analysis revealed that the epithelioid cells were positive for HMB-45 and smooth muscle actin. Furthermore, the atypical epithelioid cells displayed P53 immunoreactivity and mutation at exon 7 for p53. The tumor showed a typical monoclonal pattern but no loss of heterozygosity or microsatellite instability. Markedly atypical epithelioid cells with vascular invasion, distant metastasis, and fatal outcome were interpreted as malignant characteristics of hepatic AML. It is suggested that large tumor size, pleomorphic nuclei with high proliferation activity, and P53 immunoreactivity may predict the existence of malignant transformation of hepatic AML.

A rapid and simple analytical method was developed for the simultaneous and quantitative determination and separation of hydrophilic imidazolium ionic liquids (ILs) (1-butyl-3-methylimidazolium chloride, [C(4)mim]Cl; 1-hexyl-3-methylimidazolium chloride, [C(6)mim]Cl; 1-octyl-3-methylimidazolium chloride, [C(8)mim]Cl; 1-allyl-3-methylimidazolium chloride, [Amim]Cl; or 1-allyl-3-methylimidazolium bromide, [Amim]Br) with miscible ethyl acetate and EtOH and their mixtures using reverse phase liquid chromatography coupled with refractive index detection (RPLC-RI). The influence of 60 to 100% (volume percentage) methanol in the mobile phase on the IL systems ([C(4)mim]Cl, [C(6)mim]Cl, [C(8)mim]Cl, [Amim]Br, or [Amim]Cl)-ethyl acetate-EtOH was investigated. The optimum mobile phase for the system [C(8)mim]Cl-ethyl acetate-EtOH, [C(4)mim]Cl-ethyl acetate-EtOH, [Amim]Br-ethyl acetate-EtOH and [Amim]Cl-ethyl acetate-EtOH was methanol/water (60:40, v/v), and methanol/water (70:30, v/v) for [C(6)mim]Cl-ethyl acetate-EtOH. Under optimum mobile phase conditions for each system, the RSD of the retention time ranged from 0.02 to 0.04%, and the RSDs of the peak area percent ranged from 0.23 to 1.85%, which showed good reproducibility of the RPLC-RI method. The RPLC-RI method can determine IL, ethyl acetate, and EtOH simultaneously in 5 min, and the analytes, especially IL, can be eluted completely. The results show that the RPLC-RI method can be used to separate and determine ILs in mixtures with organic compounds simultaneously and quantitatively.

Arabinogalactan derivatives conjugated with gadolinium-diethylenetriaminepentaacetic acid (Gd-DTPA) by ethylenediamine (Gd-DTPA-CMAG-A2) or hexylamine (Gd-DTPA-CMAG-A6) have been synthesized and characterized by means of Fourier transform infrared spectra (FTIR), 13C nuclear magnetic resonance (13C NMR), size exclusion chromatography (SEC), and inductively coupled plasma atomic emission spectrometry (ICP-AES). Relaxivity studies showed that arabinogalactan-bound complexes possessed higher relaxation effectiveness compared with the clinically used Gd-DTPA, and the influence of the spacer arm lengths on the T1 relaxivities was studied. Their stability was investigated by competition study with Ca2+, EDTA, and DTPA. MR imaging of Wistar rats showed remarkable enhancement in rat liver and kidney after i.v. injection of Gd-DTPA-CMAG-A2 (0.079+/-0.002 mmol/kg Gd3+): The mean percentage enhancement of the liver parenchyma and kidney was 38.7+/-6.4% and 69.4+/-4.4% at 10-30 min. Our preliminary in vivo and in vitro study indicates that the arabinogalactan-bound complexes are potential liver-specific contrast agents for MRI.
